Cleaning slides

Dried photographic emulsion is removed prior to imaging. We use a combination of isopropanol based lens cleaning solution mixed with 1% NaOH. This solution dissolves NBT2 emulsion on the back of the microscope slide and removes dust and debris that settled on the slides during coverslipping or drying.

Photography

 

1. Equipment:
   
a.
Images are captured using a Zeiss Stemi 11 Stereomicroscope set at low magnification sufficient to capture the entire section of coronal and saggital tissues.
   
b.
Darkfield images are illuminated using a 3” MVI edge illuminator connected to a 15 W halogen light power source set on maximum for all photography. Reference cresyl violet images are captured using the brightfield settings on the microscope.
 
c.

Images are captured using a Photometrics Coolsnap es Monochrome ADC 20 Mhz Camera made by Sony with a 1300 x 1040 High resolution Interline chip directly mounted to the Zeiss Stemi 11 Stereomicroscope.

 
d.
MetaView (Version 6.4a) software is used to control the camera and to properly orient each image for display on the website

Darkfield images are captured and the exposure time of capture using the Metaview software is logged into our project management database. Exposure time varies for each microscope slide depending on the signal strength. We attempt to capture a field identical to what is observed on the slide through the eyepieces of the microscope. Image capture time ranges from <30 ms to >130 ms.
 
f.
Images are captured as grayscale .TIFF files and converted to .JPEG files for permanent storage.
2. Image processing: Images are imported into Adobe Photoshop 7.0 to optimize brightness, contrast and gamma levels and to remove debris from the background using the healing brush tool.
3. Images are then converted to intermediate and thumbnail sizes using Adobe Photoshop 7.0. for display on the website:

Full size: 1392 x 1040 pixels 2

Intermediate size: 500 x 328 pixels 2

Thumbnail size: 200 x 130 pixels 2

4. Image upload: All images are uploaded to a holding folder on the server for display on the website. Images are uploaded to the website server using FTP Voyager software version 10.0 by RhinoSoft.com ( Helenville , WI , 53137 ).
